Fused Pyrimidine Heterocycles - Dihydrofolate Reductase Inhibitors. 

Folates are crucial for cell replication as they take part in biosynthesis of nucleotides by donation of C5 methyl group of dTMP, C2 and C8 ring atoms of purine nucleotides. Essential role of dihydrofolate reductase enzymes (DHFR) in the control of intracellular concentrations of nucleotides makes them a perfect target to achieve antitumor, antimicrobial and antiprotozoal therapeutic effects. Several folate cofactor analogues have found use in the clinic as antibacterials and anticancer drugs. In the report a brief review on the known and most potent DHFR inhibitors will be presented. Synthetic strategy for the synthesis of novel thieno[2,3-d]pyrimidines as potential DHFR inhibitors and results on their DHFR inhibitory activity will be dicussed, as well.

 Palladium-Catalyzed Cross-Coupling Reactions in a Pyrimidine and Pyrrolopyrimidine Series. 

Nitrogen heterocycles with alkynyl and aryl substituents are high value synthetic targets and can be found as substructures in numerous pharmaceutical and biological active compounds. They are of great interest as electron transporting materials in OLED devices. For the synthesis of alkynyl and arylpyrimidines and pyrrolopyrimidines the direct alkynylation and arylation is a simple approach that does not involve the preparation of specific intermediates needed in the synthesis via cyclisation procedures. Some our own results on the palladium catalysed cross-coupling reactions of halopyrimidines and pyrrolo[2,3-d]pyrimidines with terminal alkynes and arylboronic acids will be presented. Influence of substituents, catalyst, ligands and other reaction conditions will be discussed.
